Nació el 17 de agosto de 1932 en Pessac, Francia. A los 18 años se instaló en París y en 1951 publicó su primer trabajo en el semanario Sud Ouest Dimanche. En aquellos tiempos conoció a René Goscinny, y crearon Las aventuras del Pequeño Nicolás, en 1955 en formato de historietas semanales. Luego, se convirtieron en textos ilustrados para Sud Ouest Dimanche. Estos relatos fueron compilados en una colección de libros de enorme éxito mundial. En paralelo, Sempé colaboró con sus cartoons para la revista Paris Match y otras publicaciones. Su primer libro de humor gráfico, Nada es simple, se publicó en 1962. A este le siguieron Todo se complica, El gran pánico y muchos más. Sus cartoons comenzaron también a aparecer fuera de Francia en revistas como la inglesa Punch y la estadounidense Esquire. Y en 1978 inició su colaboración con la mítica revista The New Yorker: sus impactantes portadas exhiben a la perfección su sensibilidad y su talento.
Sempé es considerado un verdadero maestro del dibujo humorístico, un poeta que dejaba ver, a través de su sofisticado ?y a la vez tan sencillo? humor, todas las bondades y defectos del ser humano. Falleció en Francia el 11 de agosto de 2022.
